Formula and methodology
Formula: Observed mutation frequency = mutants observed ÷ total tested; percent = frequency × 100; per-million frequency = frequency × 1,000,000.
Mutation frequency is a proportion of the tested population carrying or expressing the measured mutant outcome. Reporting the raw fraction, percent, and per-million form makes the denominator visible and keeps frequency distinct from a time-normalized mutation rate.

Assumptions and limits
- The mutant count is part of the total tested population and both counts use the same sampling frame.
- The calculation is an observed frequency; it does not estimate a de novo mutation rate, correct for clonal expansion, or model detection bias.
- A count of zero is valid and produces zero frequency, while a mutant count above the tested total is rejected as an inconsistent input.
